About Color replacer effect and good way to make new layer.

Hello. I have 2 questions.
About color replacer effect, is it possible to change only 1 specific color using this effect?
Problem is if layer has similar colors, color replacer change them all, something like,
Layer has 2 colors
Color A = R 255 G 0 B 0
Color B = R 254 G 0 B 0
in this case I only want to change Color A to R 0 G 255 B 0
so I pick Color A in Fx stack and apply the effect, but it changes not only Color A but also Color B. I don't want to change Color B.
And 2nd question.After I finish drawing line layers, I make at least 2 to 3 color layers for each line layers, but is it possible to make new empty layers that have a same exposure structure of line art? I mean for example, there is line layer that has 3 frames and has 3 instances for each frames, so I need to make new layers that is same length, has 3 empty instances to do painting on them.
If line layer is much longer and has complicated exposure structure, to make new layers for painting will be a pain.
Please give me your advice.
